Aromatic-cationic tetrapeptide D-Arg-Dmt-Lys-Phe-NH2 (elamipretide)
SS-31 is a synthetic tetrapeptide, D-Arg-Dmt-Lys-Phe-NH2, where Dmt is 2',6'-dimethyl-L-tyrosine. It carries alternating cationic and aromatic residues, a D-amino acid at position one and a C-terminal amide. The compound is one of the Szeto-Schiller (SS) peptides designed by Hazel Szeto and Peter Schiller. Its nonproprietary name is elamipretide, and it was earlier known by the development codes Bendavia and MTP-131.
SS-31 is classed as a cell-permeable aromatic-cationic peptide. Published work spans structure-activity comparisons across related tetrapeptides, lipid-bilayer binding biophysics, protein-interaction mapping by cross-linking mass spectrometry, and molecular-dynamics simulation. The references below are limited to these chemistry and biophysics papers.
